Abstract

Kutatásunk során a lézeres szórási interferencián (laser speckle) alapuló véráramlásmérésre egy olyan eljárást dolgoztunk ki melynek pontossága (különösképpen bőrszövet esetén) jóval meghaladja a hagyományos, egyetlen expozíciós idő alkalmazásán alapuló módszerét. Az első szakaszban a bőrszövetet egy modellel helyettesítettük, melyben a vörösvértestek mozgását polisztirén mikrogömb szuszpenzió áramlása, a bőr felületének fényszórását pedig a szuszpenzió felett elhelyezett áttetsző, ugyanakkor a fényt nagymértékben szóró rétegek helyettesítették. Az interferencia képét rögzítő kamerának az expozíciós idejét 0,2 ms – 500 ms tartományon való változtatásával felvettünk egy interferencia kép kontraszt (K)-expozíciós idő (T) görbét. Az ideális esetre (nincs felületi szórás) számolt elméleti K(T) függvény egy módosításának segítségével függvényillesztéssel ki tudtuk számolni az áramlási sebességre jellemző, ún. korrelációs időt, mely függetlennek bizonyult a felület szórási tulajdonságaitól és az aktuális optikai beállításoktól. A mérési eljárást teszteltük valós, bőrszöveti véráramlás (perfúzió) mérésekkel, miközben referenciamérésként a már széles körben alkalmazott lézer-Doppleres módszert alkalmaztuk. A vizsgálatok azt mutatták, hogy a különböző stimulusok hatására bekövetkező relatív perfúzió változásnak a két eljárással mért értéke jól egyezik. A felvételek rögzítési módjának optimalizálásával egy ~1 s feloldású, valós idejű mérési módszer lehetőségét mutattuk ki. | In the framework of the project a new method was developed for the laser speckle contrast analysis (LASCA) based blood perfusion measurement, which gives more accurate results as compared to the classical method based on images recorded with single exposure time value. As the first step a skin model was built using polystyrene microsphere suspension covered with semitransparent strongly scattering layers. The exposure time of the camera which recorded the speckle patterns was varied in 0.2 ms- 500 ms range, and a speckle contrast (K) –exposure time (T) curve obtained. By modifying the theoretical K(T) function derived for ideal case (when scattering from the skin surface is neglected) and fitting it to the measured data the correlation time related to the average flow velocity was determined and found to be independent of the scattering properties of the surface and the optical settings. The measurement method was tested with skin perfusion measurements, where data obtained from the well established laser-Doppler measurements serves as reference. It was found that the relative perfusion change caused by ischemia and measured with the two method are in good agreement. With optimization of the image recording, real-time monitoring with ~1 s temporal resolution can be achieved.
