Abstract

Munkánk során humán korneális epitélsejteken és keratinocitákon vizsgáltuk az intracelluláris mintázatfelismerő Nod-like receptor (NLR) család expresszióját és a fehérjék lehetséges szerepét. Kimutattuk, hogy (1) UV-B sugárzás hatására az NLR tagok expressziója csökken; (2) a korneális epitél sejtvonal (HCE-T) nem expresszálja az inflammoszoma ASC adaptor molekuláját és feltehetően ez a magyarázzata annak, hogy ezek a sejtek nem képesek IL-1beta termelésre; (3) a Nod2-szignaloszoma, amely főként a bakteriális fertőzések elleni védekezésben vesz részt, jól működik a sejtvonalban; (4) limbális őssejtekből (LSC) differenciáltatott epitél sejtek expresszálják az NLR-eket; (5) ezek gén szintű expressziója összevethető a fotorefraktív keratektómiából (PRK) származó primer korneális epitél sejtek NLR expressziójával; (6) LSC sejteken beállítottuk a Pseudomonas aeruginosa fertőzést az NLR-ek működésének további vizsgálataihoz; (7) humán primer keratinocitákon a TLR agonisták közül egyedül a duplaszálú RNS-t mimikáló polyIC képes IL-1beta termelést indukálni; (8) a polyIC indukálja a Nalp3 inflammoszoma tagok expresszióját, fokozza a kaszpáz-1 enzim aktivitását, továbbá kaszpáz-1 inhibitorral gátolható az IL-1beta termelés; (9) polyIC kezelés hatására indukálódik a Nod2 expressziója és ezt követően a Nod2 specifikus ligand (MDP) kezelés szinergista módon fokozza a gyulladásos citokinek (IL-6, IL-8, TNF) és az antimikrobiális peptidek (beta-defensin, CAMP) expresszióját. | In our work, we studied the expression and function of the intracellular pattern recognition receptor family, Nod-like receptors (NLRs) in human corneal epithelial cells and in keratinocytes. We found that (1) the expression of NLRs is decreased by UV-B irradiation; (2) the corneal epithelial cell line (HCE-T) fails to express the inflammasome adaptor ASC which results in the loss of IL-1beta expression in these cells; (3) the Nod2-signalosome, which participates in the responses against bacterial infections, functions properly in this cell line; (4) limbal stem cell (LSC) derived corneal epithelial cells express NLRs; (5) the expression level of NLRs from LSC is comparable with the NLR expression from photorefractive ceratectomy (PRK); (6) we established the Pseudomonas aeruginosa infection on LSCs to study the NLR function; (7) on human primary keratinocytes only the double stranded RNA-mimicking polyIC is able to induce the secretion of IL1-beta; (8) polyIC induces the expression of Nalp3 inflammosome components, induces caspase-1 enzymatic activity and caspase-1 inhibitor reduces IL-1beta production; (9) polyIC induces the expression of Nod2 and subsequent treatment with Nod2-specific agonist (MDP) synergistically induces the production of inflammatory cytokines (IL6, IL8, TNF) and antimicribial peptides (beta-defensin, CAMP).
