Abstract

Staphylococcusok a fő kórokozói a nozokomiális fertőzéseknek. A poli-béta-(1-6)-N-acetil glükózamin (PNAG) sejtfelszíni poliszacharid antigén a Staphylococcus aureus és S. epidermidis fertőzések egyik virulens faktora és ezen baktériumok biofilm mátrixának fő komponense. Először szintetikus utat dolgoztunk ki 2-ftálimido csoporttal rendelkező tioglükozidok előállítására, amelyek ideiglenes védőcsoportként klóracetil csoportot, valamint állandó védőcsoportként acetil csoport tartalmaztak. Ezeket a kulcs intermediereket brómozással donorrá, valamint a terminális primer hidroxil csoport szelektív felszabadításával akceptorrá alakítottuk. A nagyobb tagszámú oligoszacharidok előállítására blokkszintéziseket alkalmaztunk. Leírtuk a béta-(1-6) interglikozidos kötés tartalmazó, valamint feniltio aglikonnal rendelkező N-acetil-glükózamin oligoszacharid sorozat szintézisét, melyekkel elvégeztük a staphylococcusok biofilmjét oldó képességgel rendelkező Dispersin B enzim szubsztrát specifitásának a vizsgálatát. Res. 346, 1445, (2011)) A BSA-hoz történő konjugálási lépés tanulmányozásához előállítottunk formil-heptil aglikonnal rendelkező maltóz származékot, majd elvégeztük a kapott glikokonjugátumok immunológiai vizsgálatát. (Electrophoresis, közlésre elfogadva) Megvalósítottuk a PNGA di-, tetra-, hexaszacharid fragmensének a szintézisét formil-heptil aglikonnal, majd az oligoszacharidokat BSA-hoz konjugáltuk reduktív aminálással. (Glycoconjugate J., közlésre benyújtva) | Staphylococci are major causes of nosocomial infections. Poly-beta-(1-6)-N-acetyl glucosamine surface polysaccharide antigen (PNAG) is a virulence factor for both S. aureus and S. epidermidis infections and a major component of the biofilm matrix of these pathogens. First, we have developed a synthetic route for the synthesis of 2-phthalimido-containing thioglucosides equipped with monochloroacetate ester as temporary protecting group and acetate esters as permanent protecting groups. These key-intermediates were transformed either to a donor by bromination or to an acceptor by selective liberation of the terminal primary hydroxyl group. We have used blocksyntheses for the preparation of higher membered oligosaccharides. We have described the synthesis of beta-(1-6)-linked N-acetyl-glucosamine oligosaccharide series with phenylthio aglycone (from disaccharide to pentasaccharide) and their application to investigate the substrate specificity of Dispersin B enzyme which exhibits staphylococcal biofilm detachment activity. Res. 346, 1445-1453, (2011)) We have realized the synthesis of maltose bearing formyl heptyl aglycone to investigate the conjugation process to BSA by reductive amination and the immunogenecity of the obtained neoglycoproteins. (Electrophoresis, in press) We have achieved the preparation of di-, tetra- and hexasaccharide fragments of PNAG with formyl-heptyl aglycone and their attachment to BSA by reductive amination.
