Abstract

A posztdoktori OTKA projekt fő célja szabályozott és elnyújtott hatóanyag-leadó rendszerek fejlesztése volt, melyeket aminosavakból felépülő multireszponzív polimer gélekből valósítottunk meg. A poliaszparaginsav alapváz pH érzékenységét a keresztkötő vagy a másik polimer alkotó hőmérséklet-, redoxpotenciál- vagy enzim érzékenységével kapcsoltuk össze. Analitikai módszereket dolgoztunk ki, fejlesztettünk tovább, polimer gélek tulajdonságainak jellemzésére. Hatóanyag- és egyéb modellvegyületek szabályozott kioldódását számos paraméter függvényében részletesen jellemeztük. Biológiai vizsgálatok alapján a szintetizált gélek ígéretesek, jövőbeli orvosbiológiai alkalmazásuk lehetséges, elsősorban gasztrointesztinális hatóanyag-leadó rendszerként és implantátumként, vagy olyan esetekben, ahol az in situ gélesedés előnyös. A pályázathoz közvetlenül kapcsolhatóan 3 db közleményünk jelent meg nemzetközi folyóiratban és további 3 db kézirat készül jelenleg. Hazai és nemzetközi konferenciákon 7 db előadást és 14 db posztert mutattunk be a pályázati időszak alatt. | The main aim of OTKA postdoctoral project was developing controlled and prolonged drug delivery systems. Our particular attention has been paid to multiresponsive amino acid based polymers and hydrogels in order to achieve more speci?c answers to environmental triggers and to approach the complex behaviour of living systems. Intelligent polymer gels were synthesized with the combination of pH responsive poly(aspartic acid) and the temperature, redox potential or enzyme sensitive cross-linker or copolymer. Analytical methods were developed for characterization of the polymer gels. The controlled delivery of molecular payloads and drugs were studied in detail with a variation in parameters. The key issues such as material processability, stability, biodegradability, toxicity and efficient controlled release of molecular payloads were studied. The stimuli sensitive properties along with the high water content and good mechanical stability make our hydrogels good candidates for human biological applications such as drug delivery systems and implants, as well as for further applications in which in situ gelation is bene?cial. The scientific results were demonstrated in 3 pieces of papers in peer-reviewed journals. 7 pieces of oral and 14 pieces of poster presentations were published in Hungarian and international conferences.
