Abstract

Munkánk során kimutattuk, kismólsúlyú hősokk-fehérjék (sHSP-k) kardioprotektív hatással rendelkeznek transzgenikus egér modellen. A hsp27 fehérjét overexpresszáló egérvonalakban a tumorellenes szerrel (doxorubicin) történő kezelés káros mellékhatása jelentősen csökkent. A kvantitatív RT-PCR kísérletek során nyert adatok arra utalnak, hogy az sHSP-t overexpresszáló sejtekben a károsodott fehérjék lebontását végző gének kifejeződése csökken, ami erősen alátámasztja az sHSP-k citoprotektív szerepét. Kísérleteink rávilágítottak arra is, hogy a stressz-indukált HSP expresszióban kulcsszerepet játszhat a membránkötött Rac1 által mediált jelátviteli mechanizmus is. Ezen eredmények elősegíthetik új, membránaktív gyógyszerek fejlesztését is. Az sHSP-k védőszerepét szerepét demonstráltuk hasadó élesztő (S. pombe) esetében is. Ezen „micromammal”-nak is nevezett modelszervezetben kimutattuk, hogy a két sHSP eliminációja –a membránvédő szereppel felruházott trehalóz jelenlétének ellenére - befolyásolja sejtmembrán összetételét, előrevetítvén modellünk alkalmazhatóságát a gyógyszerfejlesztés terén is. | We have shown that small heat shock proteins (sHSPs) have cardioprotective effects in transgenic mouse model. Mice overexpressing hsp27 and treated with the anti-tumor agent (doxorubicin) showed significantly reduced adverse side effects compared to wild type mice. Quantitative RT-PCR data showed that in the sHSP-overexpressing cells the expression of genes involved in protein degradation has been significantly reduced, strongly supporting the cytoprotective role of sHSPs .Our experiments revealed that membrane bound Rac1 might play a key role in mediating the expression of HSPs These results may facilitate the development of new membrane–active drugs. We have also demonstrated the protective role of sHSPs using the fission yeast (S. pombe) as a model. We have shown that elimination of the two sHSPs (despite the presence of trehalose) in the ""micromammal"" affect cell membrane composition, anticipating the applicability of the model in the field of drug discovery as well.
