Abstract

A szalicilsav (SA) a paradicsom sóstressz akklimatizációját javító hatását korábban megvilágítottuk. Bár ezt kapcsolatba hozták a fotoszintézis hatékonyságának fokozódásával és az antioxidáns mechanizmusok aktiválódásával, megállapítottuk, hogy az e szempontból azonosan ható 10-7 és 10-4 M-os SA előkezelések közül csak a nagyobb koncentráció hatásos. Eközben a kontrollal azonos fotoszintetikus aktivitás mellett oldható cukrok akkumulálódnak, ozmotikus adaptáció történik a 10-4 M-os SA előkezelésnél, ami sókezelés mellett is megmarad. A sóstressz alatt az előkezelt növényekben megnövekedett fotoszintetikus aktivitás mérhető. Átmeneti oxidatív stressz után az előkezelés végén magasabb abszcizinsav (ABS) szinteket és különösen a gyökerekben magasabb aszkorbinsav és glutation mennyiségeket mértünk a magasabb SA koncentrációnál. Az ABS szintézis fokozódását a 10-4 M-os SA a zeaxanthin epoxidáz és egyes abszcizinaldehid oxidáz izoenzimek, míg az ABS a 9-cisz epoxikarotinoid dioxigenáz expressziójának fokozásával éri el. A sóstressz ebben az esetben egy ozmotikusan adaptálódott, magas ABS és nem enzimatikus antioxidáns tartalmú gyökérszövetet ér, ami előnyt jelent a kontroll állapothoz képest. A sikeres akklimatizációt mutatja a detoxifikációban szerepet játszó glutation transzferáz gének expressziójának növekedése is. A SA a sejtek programozott halálát elsősorban a reaktív oxigén formák (ROS) mennyiségének fokozásával, míg a sóstressz az etilén által indukált ROS növelésével éri el. | Our earlier results revealed the beneficial effect of salicylic acid (SA) on the acclimation of tomato to high salinity. It was suggested that SA effect was associated with enhanced photosynthesis and activation of the antioxidant system. However, both 10-7 and 10-4 M SA improved these physiological parameters but only 10-4 M SA could mitigate the salt stress injury. During „chemical hardening” the plants maintained photosynthetic activity at control level and became osmotically adapted by the accumulation of soluble sugars. This status was preserved under salt stress. Plants exposed to 10-4 M SA displayed the symptoms of transient oxidative stress, and accumulated abscisic acid (ABA) and non-enzymatic antioxidants, ascorbic acid and glutathione in the roots. The biosynthesis of ABA was controlled by SA by the increased expression of zeaxanthin epoxidase and abscisic aldehyde oxidases, while ABA enhanced the expression of 9’-cis-epoxicarotenoid dioxigenase autocatalitically. The root tissues exposed to 10-4 M SA have already been osmotically adapted, contained higher level of ABA and non-enzymatic antioxidants when they were exposed to salt stress. This ensures advantage for hardened plants compared to controls. The increased expression of glutathione transferases also contributes to successful acclimation of plants. Lethal concentrations of SA induced the death of cells (PCD) by the generation of ROS, while ethylene-generated ROS is a component of salt stress-induced PCD.
