Abstract

A Fusarium verticillioides a kukorica egyik fontos kórokozója, ismert mikotoxin-termelő penészgomba. Az egészségügyi kockázatot okozó fumonizinek poliketidekből képződő hasonló szerkezetű mikotoxinok. A fumonizineket a szénlánc elején lévő funkciós csoportok szerint négy fő csoportba (A, B, C, P) sorolják. Mikotoxikológiai szempontból a B-csoportba sorolt fumonizinek (FB1, FB2, FB3, FB4) a legjelentősebbek. Élelmiszer biztonsági szempontból az FB1 a legfontosabb, amely az FB analógok 70-80%-át képviseli. A különböző fumonizin-csoportba tartozó analógokat nehéz kimutatni a hagyományos elválasztás-technikai eljárásokkal. Az általunk használt érzékeny, szelektív módszerek (RP-HPLC/ESI-TOFMS, RP-HPLC/ESI-IT-MS) esetében csökkent a műtermék képződés lehetősége, valamint a gombatenyészetekből készített kivonatok további tisztítás nélkül használhatók eddig ismeretlen fumonizin analógok kimutatására azonosítására. Optimizáltuk az RP-HPLC elválasztás, valamint a F. verticillioides tenyészetekből történő fumonizinek kivonásának paramétereit. Vizsgálataink során többek között 28 FB1 izomert, új FB5 izomereket, észterezett FB1 komponenseket azonosítottunk. Hatvan hazai F. verticillioides törzs fumonizin-termelésének vizsgálata során három kemotípus létezését bizonyítottuk. PCR módszert alkalmaztunk fuzárium-fajok és fumonizin-termelő fuzárium-törzsek azonosítására. Fumonizin-termelő F. verticillioides törzsek dimorfizmusát bizonyítottuk. Folyamatban van a moniliformin kimutatása. | The fumonisins are a group of polyketide-derived, structurally related mycotoxins producing mainly by Fusarium verticillioides, which can cause disease in plants and animals. The fumonisin analogs can classified into four main groups, identified as the fumonisin series A, B, C and P. The fumonisin B (FB) analogs, comprising toxicologically important FB1 is most abundant naturally occurring fumonisins. The lesser known fumonisin analogs are difficult to detect with most analytical techniques, but they can be analyzed by RP-HPLC/ESI-TOFMS and RP_HPLC/ESI-IT-MS. The detection of 28 FB1 isomers, new FB5 isomers and esterified FB1 toxins yield further data for a more detailed knowledge of their biosynthesis. Our results demonstrate the amount of the examined new fumonisins and fumonisin-like compounds is 20 percents of the described fumonisins, which is a remarkable amount to consider their potential toxicity. According to our results we have identified naturally occuring variants (chemotypes) of fumonisin and moniliformin producing isolates of F. verticillioides. PCR analysis with species-specific primers confirmed morphological identification of F. verticillioideas and other Fusaria. The VERT1/2 set of primers were used to discriminate potential fumonisin producing strain of F. verticillioides. A study was carried out on the distributed and diversity of Fusarium species associated with non-agricultural grasses in Hungary. The yeast-like form of F. verticillioides is discussed.
