Abstract

A pályázat támogatásával célunk olyan többszörösen transzgenikus zebradánió vonalak létrehozása volt, amelyek a fő toxikológiai célszervekben (máj, érszövet, vér, idegrendszer) fluoreszcens fehérjéket termelnek, így lehetővé teszik a mérgező anyagok hatásának gyors, in vivo detektálását és nyomon követését a jelölt szervekben. A munka során létrehoztunk egy új génkonstrukciót, amely a májban ösztrogénnel indukálható fluoreszcens fehérje kifejeződését eredményezi. A konstrukciót vad típusú és transzgenikus zebradánió embriókba injektáltuk és néhány alapító egyedből kiindulva létrehoztuk a transzgenikus vonalak F1 majd F2 generációját. A gén konstrukció integrációs helyének és kópiaszámának meghatározását megkezdtük, de költségtakarékossági okokból, csak az ösztrogén indukció alapján a legmegfelelőbb kiválasztott vonalakon végezzük el. A vérben, erekben és az idegrendszerben fluoreszcens fehérjéket kifejező transzgenikus vonalakból kétszeresen transzgenikus, homozigóta F2 vonalakat, valamint többszörösen transzgenikus hemizigóta egyedeket hoztunk létre. Megállapítottuk, hogy a kék színű fluoreszcens fehérje (EBFP) nem alkalmas az indukálható, májspecifikus konstrukció létrehozásához az általunk felállított vizsgálati rendszerben, ezért egy új fluoreszcens fehérje bevonásával (CFP) megkezdtük egy újabb májspecifikus konstrukció kialakítását. | In the framework of the project our aim was to develop multiple transgenic lines expressing fluorescent proteins in the toxicologically important target organs (liver, blood vessels, blood cells, nervous system). These newly developed lines would enable the fast in vivo detection of toxic effects on the labelled organs and the registration of changes. During the project we developed a new transgene construct that drives inducible fluorescent protein expression in the liver. This construct was injected to 1-2 cell wild-type and transgenic zebrafish embryos and the F1 and F2 generation of 3 founders was established. The determination of the transgene integration site and copy number is in progress, however, in order to save cost, it will be performed only on individuals showing the highest fluorescent signal inducibility as a response to estrogenic treatment. In the framework of this project, double transgenic, homozygous F2 individuals and hemizygous, multiple transgenic fish were produced from the available single transgenic lines in which the blood cells, the blood vessels or the nervous tissues are fluorescently labelled. We found out, that the EBFP is not a suitable reporter for developing such transgenic lines, as the signal it produces is too weak to be detectable. So a new blue fluorescent protein (CFP) was selected for building a new, blue construct. Purchasing the encoding plasmid caused a delay.
