Abstract

Antidepresszánsokkal MT gátolt és NAT-KO egereken igazoltuk, hogy az extracelluláris MA koncentráció jelentős tényező az immunmodulációban. Akutan az antidepresszánsok gyulladásgátlók, krónikusan azonban nem, azaz sokkal komplexebben, több célponton hatnak. Antidepresszánsok hatására az endotoxin-indukált plazma TNF-a szint szignifikánsan csökken, míg hippokampuszban a drogok immunmoduláns hatása más, szignifikánsan kisebb. Ennek magyarázata az adrenoceptorok megváltozott szenzitivitása ill. a drogoknak az MDR fehérjék által előidézett csökkent mennyisége lehet, amelyet CNS-t is érintő malignus betegségekben is észleltünk. Különbséget mutattunk ki NAT-KO és WT egerekben a periférián és a hippokampuszban észlelhető, LPS-indukált TNF-a szint antidepresszánsal történő modulációjában, és a c-fos és pCREB transzkripciós faktorok aktiválhatóságában is. Új szabályozó mechanizmust ismertünk fel; a stimulus függő adrenerg immunmodulációt. Igazoltuk, hogy ebben tranziens MAP-kináz foszforiláción át a b-adrenerg receptor nemcsak Gs, hanem Gi fehérjéhez is képes kötődni így elsőként mutattuk ki, hogy az immunrendszerben is létrejöhet az eddig csak más szövetek sejtjeiben ismert Gs/Gi átkapcsolás. Eszerint az adrenerg immunmoduláció Janus arcú, azaz az alkalmazott stimulustól függöen akár immunszuppresszáns gyulladásgátló, akár immunaktiváló gyulladásfokozó hatású is lehet. Állatkísérletes és in vitro eredményeink hozzásegítettek egyes humán megbetegedések hátterének jobb megértéséhez. | The importance of MA levels in immunomodulation was demonstrated in NET-KO and in MT blocked mice.As antidepressants were antiinflammatory in acute but not in chronic treatment they might act in a more complex way and on multiple targets.LPS-induced TNF-a level was significantly decreased due to antidepressant treatment, but immunomodulatory effect in the hippocampus was diverse and significantly lower, because of altered sensitivity of adrenergic receptors, or by decreased concentration of the drugs due to the action of MDR proteins that could also be observed in some malignant diseases affecting the CNS.Differences were demonstrated between the plasma and the hippocampus in modulation of the LPS-induced TNF-a levels by antidepressants, and in the activation of c-fos and pCREB transcription factors.A new regulatory mechanism was revealed: the stimulus-dependent adrenergic immunomodulation.This was due to the fact that b-adrenergic receptor was able to bind not only Gs but also Gi protein, via a transient MAPK phosphorylation.We were the first to recognize that Gs/Gi switch could also happen in the immune system as it was described in some other tissues earlier.So adrenergic immunomodulation is Janus faced, i.e. it might be either immunosuppressive, antiinflammatory or immunostimulant, proinflammatory, depending on the stimulus applied.The results of our animal experiments and in vitro studies contribute to a better understanding of the background of certain human diseases.
