Abstract

A klasszikus elméletekben leírt elektroforézist főleg gyenge elektromos terekben vizsgálták, ahol a részecskék vándorlási sebessége (V) egyenesen arányos a külső tér feszültségével (E). Az E növekedésével a linearitástól való eltérés egyre erősebb és elképzeléseink szerint a nem-lineáris tartományban a V a térerősség harmadik hatványával arányos. Kidolgoztuk a nem-lineáris ef új, az erős elektromos terekre és tetszőleges felületi vezetéssel rendelkező részecskékre érvényes elméletét az elektromos kettősréteg nem-lineáris polarizációjának figyelembevételével. Erős elektromos terekben megvalósuló elektroforézis mérésére alkalmas, elektromos impulzusok és nagy felbontású képelemző rendszeren alapuló új módszert fejlesztettünk ki. Részletesen vizsgáltuk polisztirol, grafit, aluminium-oxid-és biokolloidok (élesztősejt) elekroforézisét a térerősség (50-500 V/cm), részecske-méret, pH és ionerősség függvényében, ill. adszorbeált polimerek jelenlétében. Kimutattuk, hogy- ellentétben a klasszikus elmélettel- E =100 V/cm és erősebb terekben: (1) jelentős nem-lineáris effektusok figyelhetők meg és jó az összhang az elméleti és kísérleti V(E) görbék lefutása között, (2) az ef sebesség a részecskemérettel jelentősen növekszik, (3) a polimer-adszorpció csak kismértékben változtatja meg a részecskék ef sebességét. Ez azt jelenti, hogy a nem-lineáris ef független a részecskék zéta-potenciáljától és értékét a felületi vezetés és kettősréteg Dukhin (Du)-számmal jellemzett polarizációra való képessége határozza meg. | Classical theories describe the electrophoresis (ef) in weak electric fields where the electrophoretic velocity of particles (V) is proportional to the electric field gradient (E). With increasing E, the deviation from the linear V(E) proportionality increases and, according to our ideas, V is proportional to the third power of the field voltage. We have developed a new theory of non-linear ef that is valid for strong electric fields and particles with arbitrary surface conductivity, which takes into account the non-linear polarization of the electrical double layer (EDL). A new method to measure the ef velocity in strong electric fields based on the use of short electric pulses and an image analyser equipment of high resolution has been developed. We have measured in detail the electrophoresis of polystyrene, graphite, aluminium-oxide particles and biocolloids (yeast cells) as a function of the electric field gradient (50-500 V/cm), particle size, pH and ionic strength as well as vs. the adsorbed polymer amount. It has been shown that, in contrast to classical electrophoresis: (1) in electric fields higher than 100 V/cm substantial non-linear effects are observed and there is good accordance between theoretical and experimental V(E) plots, (2) the ef velocity substantially increases with the particles size, (3) the polymer adsorption only slightly changes the ef velocity of particles. This means that non-linear ef is independent of the zeta-potential of particles and it is determined by the surface conductivity and the electric double layer polarization characterized by the so-called Dukhin (Du) number.
