Abstract

.Alzheimer-kórban (AK) a neuronális degeneráció kialakulásáért a szenilis plakkokban lerakódó amyloidot beta peptidet (A beta) teszik felelőssé. Az A beta képződés az amyloid prekurzor protein (APP) kóros hasítása miatt jön létre. Munkánk során különböző pszichofarmakonok, antidepresszánsok, addiktív szerek, anesztetikumok APP metabolizmusban betöltött szerepét vizsgáltuk. Kísérleteinket in vitro primér neuron tenyészeten és/vagy in vivo patkányok akut ill krónikus kezelését követően végeztük. Mind a típusos, mind az atípusos antipszichotikumok AK-ban történő alkalmazása relative biztonságosnak tekinthető az APP metabolizmus szempontjából. Az antidepresszánsok jótékony hatása valószínűsíthető az AK terápiában. Az anesztetikumok jelentősen nem változtatták meg a corticalis APP protein és mRNS szinteket. A propofol, thiopentan, diazepam és midazolam nem fokozza az APP mennyiségét, így nem növeli az AK kialakulásának valószínűségét. Krónikus MDMA kezelés növeli a patkány corticalis APP mRNS és BACE szinteket. Ezen eredményeink felvetik a lehetőségét annak, hogy a drog abúzusnak szerepe lehet az idegsejt degeneráció létrejöttében. Mivel a cholesterol anyagcsere és az A beta kialakulás között szoros összefüggés áll fenn, vizsgáltuk a magas cholesterol diéta, a human apo-B 100 és biglycan gén overexpresszió hatását is az APP metabolizmusra. Vizsgálataink szerint ezen atherogén faktorok különbözőképpen hatnak az APP anyagcserére, és szerepük lehet az AK patogenezisében. | In Alzheimer's disease (AD), the amyloid beta peptide (A beta) depositing in senile plaques may be responsible for the neuronal degeneration. The A beta production is derived from the pathological cleavage of amyloid precursor protein (APP). For this reason, the impact of psychopharmacons, antidepressants, addictive drugs and anesthetics was investigated. In vitro or in vivo data were obtained from experiments on primary neuronal cultures or after acute or chronic treatment of rats. Both typical and atypical antipsychotics are considered to be relatively safe with respect to APP metabolism. Antidepressants are able to interfere with the APP processing, this medication might be beneficial in AD therapy. The applied anesthetics did not change cortical APP protein and mRNA concentration considerably. Results indicate that the impact of propofol, thiopental, diazepam and midazolam is negligible with regard to APP processing. Chronic MDMA treatment increases APP mRNA and BACE protein levels in the rat brain. These results point to new routes whereby abuse of this recreational drug can cause degeneration in neuronal cell. Since there is a link between the cholesterol metabolism and A beta deposition, the high-cholesterol diet and the individual or combined overexpression of the human apoB-100 and biglycan genes were also examined. These atherogenic factors could exert different effects on the processing of APP and may participate in the pathogenesis of AD.
