Abstract

1. A plazmamembrán (PM) foszfatidilinozitol 4,5-biszfoszfát (PtdInsP2) szintjének csökkentésére kifejlesztett, rapamycin függő heterodimerizációs rendszer módosításával megoldottuk a transz-Golgi foszfatidilinozitol 4-foszfát mennyiségének akut csökkentését élő sejtekben, és ezzel megnyitottuk az utat e lipid kompartment jelentőségének vizsgálata előtt. 2. Létrehoztunk egy olyan fehérje készletet, amellyel az 5-15 nm-es tartományban, perces nagyságrendben mesterséges kapcsolat hozható létre a PM és az endoplazmás retikulum (ER), a mitokondrium és az ER, valamint a mitokondrium és a PM között. Más laborokkal együttműködve megállapítottuk, hogy a módszer jól alkalmazható olyan folyamatok vizsgálatára, amelyben az említett organellumok együttműködve vesznek részt (pl. kapacitatív Ca2+ beáramlás vagy mitokondriális Ca2+ jel kialakulása). 3. A PM receptorok endocitózisának molekuláris szintű vizsgálatára új, energiatranszfer mérésre épülő módszert fejlesztettünk ki. Megállapítottuk, hogy HEK293 sejtekben a Gq fehérjét aktiváló 1-es típusú angiotenzin II és a 2C típusú szerotonin receptor internalizációját alapvetően meghatározza a PM PtdInsP2 mennyisége. Kimutattuk, hogy a PtdInsP2 szint csökkentésével kiváltható gátló hatásért a klatrin burok lefűződésének elégtelensége tehető felelőssé, míg a receporok laterális mozgása és a klatrin burok területén történő bekoncentrálódása nem bizonyult PtdInsP2 érzékenynek. 4. Létrehoztunk egy sokoldalú, nagy érzékenységű fluoreszcens képalkotó rendszert. | 1. We developed a molecular tool suitable for the acute depletion of the trans-Golgi phosphatidylinositol 4-phosphate pool in live cells by modifying the rapamycin-dependent heterodimerization system. The main advantage of the system is that it allows further investigation of the role of this inositol lipid. 2. We created a protein tool useable for formation of an artificial bridge between organelles such as plasma membrane (PM) and endoplasmic reticulum (ER), mitochondrium and PM, mitochondrium and ER in the range of 5-15 nm in live cells. Collaborating with various laboratories we found the system suitable to investigate processes, in which the mentioned organelles worked in coorporation (e.g. store operated Ca2+ influx, mitochondrial Ca2+ signal). 3. Based on energy transfer measurements we developed a new method to follow the endocytosis of PM receptors at molecular level. We found that internalization of the Gq coupled type 1 angiotensin II recepors and type 2C serotonin receptors was highly determined by the PM PtdInsP2 content in HEK293 cells. We showed that the failure of the fission of clathrin coated pits were responsible for the inhibitory effect evoked by the decrease of PM PtdInsP2 concentration, while the lateral movement of the receptors, as well as their concentration in the clathrin coated membrane were not affected by the PtdInsP2 depletion. 4. We set up a versatile, highly sensitive digital image system suitable for single cell fluorescent measurements.
