Abstract

A pályázatban azt vizsgáltuk, hogy a korábban in vitro mikrospóra szelekcióval előállított oxidatív stressz-toleráns kukorica DH vonalak rendelkeznek-e fokozott hideg és szárazságtűréssel, ill. van-e összefüggés a növények fokozott oxidatív stressztoleranciája hátterében álló megnövekedett antioxidáns kapacitásuk, valamint a hideg és szárazságtűrésük között. A hidegtűrést csírázáskor ill. kisnövény korban, míg a szárazság hatását a virágzást megelőző ill. virágzás időszakában vizsgáltuk. Csírázáskor, ill. kisnövény korban alkalmazott hidegkezelés jelentős mértékben csökkentette a kukorica csírázóképességét ill. fotoszintetikus aktivitását. Az oxidatív stressztoleráns növények közül 7 vonal hideggel szemben is magasabb toleranciát mutatott, mint a kontroll DH és a hibrid növények. Szoros korreláció mutatható ki a fiziológiai paraméterek és a növények antioxidáns kapacitása között. A szárazság hatására bekövetkező RWC és fotoszintetikus paraméterek változásai, valamint a pollen és termésfejlődés alapján 6 oxidatív stressztoleráns DH vonal szárazsággal szemben toleránsabb volt, mint a kontroll DH és a hibrid növények. A szárazságtűrés mértéke és a növények prolin ill. antioxidáns kapacitása között a korreláció kisebb volt. Ezek a vizsgálatok azt mutatják, hogy lehetséges hideg- és szárazságtűrő kukorica DH vonalak előállítása mikrospórák in vitro szelekciójával oxidatív stresszt indukáló vegyületek alkalmazásával. | The aim of the project was to characterize the cold and drought tolerance of the progeny of DH plants selected and regenerated from microspores under oxidative stress condition and to determine the relationship between the tolerance of oxidative stress and cold or drought of the selected DH lines. Seedlings and young plantlets were used for determination of cold tolerance, and the effect of drought was studied on adult, preflowering and flowering plants. Our results presented that 7 of 14 oxidative stress tolerant DH plants were also tolerant to cold, and their cold tolerance was correlated with their antioxidant capacity determined by the activities of the main antioxidant enzymes, such as superoxide dismutase, ascorbate peroxidase, glutathione reductase, catalase and glutathione-S-transferase. The drought-induced changes in the physiological parameters of leaves and in the pollen and pistil development presented that 6 oxidative stress tolerant DH plant showed higher drought tolerance than the control DH and hybrid plants. However, the correlation between physiological (RWC, photosynthesis) and biochemical (antioxidant capacity, prolin content) parameters under drought stress conditions was less evident as was found for cold. Four of the oxidative stress tolerant plants presented both drought and cold tolerance. Our investigations demonstrated that the in vitro microspore selection is a potential way of improving cold and drought tolerance of maize.
