Abstract

Jelen projekt során kifejlesztettünk egy sorozat új oligonukleotid szerkezetű telomeráz inhibitort (IC50 kevesebb mint 50 nmol) melyek egy kémiailag módosított szakaszból [(s4dU)n], és egy a telomeráz RNS ellen tervezett antiszensz szakaszból állnak (kiméra oligonukleotidok). Bizonyítottuk, hogy ezek az inhibitorok kölcsönhatásba lépnek telomeráz protein és RNS alegységével is. Megszintetizáltuk az egyik inhibitor Cy5 jelzett származékát és kimutattuk, hogy bejut a sejtbe. Egy új kromatográfiás módszert fejlesztettünk ki az erősen módosított és kiméra oligonukleotidok tisztítására is. A telomeráz inhibitor (X8AS) sejtkulturában aktívnak bizonyult 20-30 nap alatt ölve meg a sejteket, jó egyezésben az ideális telomeráz inhibitorokra javasolt citotoxikus hatás kinetikájával. Azt találtuk, hogy a kémiailag módosított szakasz önmagában [előnyösen az (s4dU)35]), amellett, hogy erős inhibitora a telomeráznak, a HIV replikáció potens és nem toxikus inhibitora is. Vizsgálva a gátlás mechanizmusát megállapítottuk, hogy nem a korábban kimutatott reverz transzkriptáz és telomeráz gátló aktivitás a felelős a HIV ellenes hatásért. Az (s4dU)35 a vírus sejtbe való belépését gátolja igen erősen (IC50 = 3 ng/ml). A telomeráz inhibitorok degradációs termékeinek citotoxikus hatását vizsgálva kiderült, hogy az s4dUMP és s4UMP tumor sejtkultúrában apoptózist indukál. | In this project we have developed a series of new oligonucleotide type telomerase inhibitor (IC50 less than 50 nmol) which are composed of a chemically modified moiety [(s4dU)n] and an antisense sequence designed against the telomerase RNA (chimeric oligonucleotides). Thus, the inhibitors are interacting with both the protein and the RNA subunit of the enzyme. We synthesized the Cy5-labeled derivative of an inhibitor molecule and proved that it can be taken up by the cell. We have developed a new chromatographic method for purification of the highly modified oligonucleotides including chimeric telomerase inhibitors. The telomerase inhibitor (X8AS) was active in cell culture killing the cells after 20-30 day treatment, in good agreement with the proposed mode of action for a telomerase inhibitor. It was found that the chemically modified part of the telomerase inhibitor [preferentially a (s4dU)35] beside its potent telomerase inhibiting activity, also inhibits the HIV replication. Studying the mode of action of the molecule it becomes clear that the earlier showed telomerase and reverse transcriptase inhibitory activity was not responsible for the antiviral activity. The (s4dU)35] inhibits the entry of the virus (IC50 = 3 ng/ml). We studied the citotoxic activity of the degradation products of telomerase inhibitors and found that s4dUMP and s4UMP induce apoptosis in tumor-cell culture.
