Abstract

The raf protooncogenes are the cellular counterparts of the v-raf oncogene expressed by a murme sarcoma virus The raf protooncogenes encode cytoplasmic serme/threonme-spectfic protein klnases which can be activated from different growth factor receptors by phosphorylatmn The mRNAs of raf protooncogenes are found m a large variety of normal adult tissues, including the central nervous system As concerns the dlstnbutmn and locahzatmn of their protem products (the raf klnases), very few data are to be found m the hterature This ~s the first detailed description of their hght m~croscoplc locahzatmn in neocort~cal and allocort~cal areas of rodents Preembeddmg immunoh~stochem~cal studies were performed on vlbratome sections from the brains of adult guinea pigs and albino rats The locahzatmns of two lsoenzymes, raf-1 kmase and B-raf lanase, were studied with the help of lsoenzyme-specfflC polyclonal antibodies Both of the antibodies detected raf proteln-hke tmmunoreactw~ty m many neurons and scattered ghal cells of the sensory neocortex, and the cmgular, pyriform, penrhmal and entorhlnal allocortlcal areas Pyramidal and non-pyramidal cells of Ammon's horn, granule cells of the dentate fascia and the large neurons m the hllar regmn were immunoreactwe, too The findings indicated that B-raf protein kmase and raf-1 klnase are present almost ubiquitously m the neurons of the investigated cortical structures The mtenslty of staining obtained w~th serial dfluttons of the antibodies indicated that the cytoplasmic concentratmn of B-raf kmase is tended to be higher than that of raf-1 klnase The present findings suggested that the raf kmases are locahzed m postsynaptlc structures, mainly m dendrites and cell bodies Their cytosohc locahzatlon and their abdlty to undergo mtracellular translocatmn during actlvatmn and phosphorylatmn raise the possibility that they play a pwotal role m the mtracellular signalling of neurons
