Abstract

A program a táplálkozás és az anyagcsere egymással szorosan összefüggő előagyi szabályozásában alapvető idegi és neurokémiai mechanizmusok eddigieknél részletesebb feltárására irányult. E többrétű célok megvalósítására komplex, integratív szemléletű és metodikájú mikroelektrofiziológiai, neurokémiai-biokémiai és magatartási vizsgálatokat folytattunk laboratóriumi patkányokban és rhesus majomban. Kutatásaink tengelyében, a korábbiakhoz hasonlóan, a táplálkozás és az energiaforgalom központi szabályozásában feltételezetten kiemelkedő jelentőségű előagyi glukóz-monitorozó (GM) idegsejt-hálózat sokrétű, differenciált jellemzése állt. Összességében közel 500 neuron extracelluláris egysejttevékenységét vezettük el laboratóriumi patkány és rhesus majom előagyban. Az irodalomban elsőként azonosítottunk komplex, endogén és exogén kémiai érzékenységű GM idegsejteket a nukleusz akkumbenszben, a mediodorzális prefrontális és az orbitofrontális kéregben, s igazoltuk e kemoszenzoros neuronok jelentőségét a homeosztázis fenntartásában. A programban feltárt új adatok további elméleti és preklinikai vizsgálatok kiinduló pontjaiként táplálkozási és metabolikus betegségek terápiáját célzó gyógyszer-targetek feltárásához vihetnek közelebb. | The program aimed to reveal so far little known neural and neurochemical mechanisms of closely interrelated forebrain regulation of feeding and metabolism. To achieve these multiple goals, complex and integrative microelectrophysiological, neurochemical-biochemical, and behavioral studies were performed in the laboratory rat and rhesus monkey. Similar to the past, multiple, differential characterization of the forebrain glucose-monitoring (GM) neuronal network, supposedly with great significance in the central control of feeding and metabolism, was put in the center of our research efforts. The extracellular single neuron activity of altogether appx. 500 neurons was recorded in the rodent and macaque forebrain. Unprecedentedly in the literature, GM neurons with complex, endogenous and exogenous chemosensitivity were identified in the nucleus accumbens, mediodorsal prefrontal and orbitofrontal cortices. Evidence for the significance of these chemosensory units in the maintenance of homeostasis was also obtained. New data revealed in this program may serve as starting points of further theoretical and preclinical investigations that aim to establish pharmacological targets of the therapy of eating and metabolic disorders.
