Abstract
1. We investigated the ultrastructure of the central nervous system of fresh water mussel after several fixation methods differing from those of our former investigations (Elekes and Zs.-Nagy, 1973), in course of which the sequence of the application of the fixatives was altered (osmium-glutaraldehyde- osmium; a mixture of glutaraldehyde and osmium in a ratio of 1 : 1), and further fixation with heavy metal (K₂Cr₂O₇) was employed between the glutaraldehyde and osmium tetroxide. In the course of the fixations, the effect of different pH values and buffers have been investigated and in some cases, uranyl acetate and phosphotungstic acid block staining, respectively, were also used. 2. Our former idea (Elekes and Zs.-Nagy, 1973) is supported by the present data that the composition of the vesicle population of fresh water mussel ganglia is heterogeneous in character, though the morphology of the different vesicle types, the organization of the vesicle matrices are fundamentally influenced by the fixation methods applied, and particularly by the sequence of the fixatives. 3. In the course of the different fixations the proportion of the clear and granular vesicles is different. The ratio of the clear vesicles is considerably higher when the OsO₄ precedes the glutaraldehyde or exerts effect together with it. 4. Comparing these with our former investigations (Elekes and Zs.-Nagy, 1973), further morphological (and possibly functional) relationship might be established between the vesicle population of the neurons of fresh water mussel and that of other in vertebrate and molluscan species.
