Abstract

According to earlier observations dogs desensitized with histamine deve loped prononnced leukocytosis, whereas suspensions of leukocytes prepa red from the blood of these animals showed a marked antihistamine effect in various biological test experiments. For two reasons this finding was unex pected : 1. It is known that reutrophil granulocytes contain a considerable amount of histamine. 2. It seems surprising that the leukocytes playing such a prominent role in defense against infection should contain a substance which according to the investigations of H alpern and al as well as to our own recent findings, leads to the generalization of the local infection. Our investigations provided evidence of the same unfavourable effect of the synthetic antihistami- nics on local infection with Staphylococci as was previously described by Halpern and co-workers in the case of infection with Salmonella typhi murium. We interpreted these findings as an inhibition effected by synthetic antihista- minics on the initial local release of histamine and some of the substances de scribed by Menkin at the site of the inflammation. Consequently it seemed desirable to check the effect of the natural antihistamine substance of the leukocytes on the pyogenic inflammation by the same methods. Essentially we had to deal with two questions : 1. If any influence will be exerted by the »natural antihistamine substance« on the local histamine liberation in pyogenic inflammation? 2. How will the microscopic picture of the inflammation be altered by this substance?
