Abstract

One of the primary bottlenecks in the industrial production of live attenuated virus vaccines is the extremely long duration of freeze-drying cycles. Since the stability of these products can only be guaranteed within a narrow temperature range, increasing efficiency requires technological optimization while strictly adhering to safe operating limits. In this research, we investigated the possibilities of cycle time reduction using a dynamic process model based on the mechanical parameters of an industrial scale unit. During the simulations, the collapse temperature (Tc) and the glass transition temperature (Tg) known from literature were considered the primary physical constraints. Our results confirm that reserves exist in empirical technological recipes, by shortening the post freezing dwell stages and implementing dynamic, near limit control of the shelf temperature profile, the primary drying time can be reduced without compromising the quality of the final product.
