Abstract
N,N- and N,O-bidentate organic difluoroboron fluorophores can be developed from natural and synthetic molecules. We report here the multistep synthesis and characterization of fluorescent boroisoquinolines as N,O-bidentate-BF2-complexes, structurally similar to cannabinoids. These compounds were envisaged to be CB1 ligands with intrinsic fluorescence that enables their use in receptor imaging without the need for an additional fluorophore. Binding and functional activity of the most promising boroisoquinoline was confirmed on CB1 receptors. This compound labeled CB1 receptors effectively in live cells allowing confocal imaging. These features nominate this compound as a new intrinsically fluorescent probe for CB1 receptor.
