Dose-dependent short-term effects of T-2 toxin exposure on some lipid peroxidation and antioxidant parameters of laying hens

Abstract

This study investigated the short-term effects of oral T-2 toxin exposition on oxidative stress parameters in different tissues in laying hens. Laying hens were fed with experimentally T-2 contaminated feed for 12 hours in 4 treatment groups (control, 5, 10 and 15 mg T-2 toxin/kg feed). After 4 and 8 hours of toxin-exposure, blood samples were taken, and at 12 hours birds were killed and blood plasma, liver, spleen and kidney samples were taken. Blood plasma malondialdehyde concentration decreased as function of time in all T-2 toxin loaded groups, and it was significantly lower in the group fed with 15 mg T-2 toxin/kg feed as compared to the control. Reduced glutathione concentration and glutathione peroxidase activity in blood plasma did not show time- and dose-dependent changes. Conjugated diene and triene levels were significantly higher at the end of the trial in the 15 mg T-2 toxin/kg feed treatment in liver. However, malondialdehyde content did not change in the tissue homogenates. Reduced glutathione concentration and glutathione peroxidase activity were significantly higher at the highest dose T-2 toxin in liver homogenates, but not in spleen or kidney. The results suggest that short-term oral exposure of T-2 toxin initiates lipid oxidation in the liver at the highest dose applied, but the antioxidant defence system eliminates the free radicals, thus the process did not terminate.
