Abstract

Új eredmények 1. A szisztémás lupus erythematosus (SLE) betegségben szenvedő betegek T limfocitáiban a protein kináz C (PKC) izoenzimek közül a béta, delta, az éta, az epszilon, a théta és a zéta, a monocitákban pedig a PKC delta, epszilon és zéta sejten belüli expressziója mind fehérje, mind hírvivő RNS szinten csökkent. Ez SLE specifikus jelenség, és a kimutatott defektus érvényesül ezen fehérjék géntranszkripciójának a szintjén. 2. A glükokortikoszteroid kezelés hatására ezek mennyisége - a T sejtekben található PKC théta kivételével -megemelkedik. 3. Az SLE-s betegek monocitáiban található csökkent arachidonsav termelés a PKC delta alulműködéséhez köthető, ami új szempontot jelent ezen sejtek SLE-ben megfigyelt kóros működésében. 4. Az SLE-s betegekben a CD4+ CD25+ Fox P3+ regulatórikus (szupresszor) T sejtek abszolut száma csökken a perifériás vérben, ami plazmaferezis hatására - a betegek javulásával együtt -megemelkedik. 5. HaCaT keratinocitákban kimutattuk, hogy a PKC alfa és PKC delta serkenti a differenciálódást és apoptózist, míg ezzel párhuzamban gátolja a proliferációt és tumor növekedést immundeficiens egerekben. Ezzel ellentétben a PKC béta és epszilon in vitro és in vivo is növeli a keratinociták sejtosztódását és gátolja a differenciálódást és az apoptózist. | NEW RESULTS 1. In the patients with systemic lupus erythematosus (SLE) the decreased expressions of some protein kinase C (PKC) isoenzymes can be observed at both protein and messanger RNA levels as follows, in the T cells, PKC beta, delta, eta, epsilon, theta, zeta; in the monocytes PKC delta, epsilon and zeta. This phenomenon is SLE specific, and it reflects the defects in the synthesis of these proteins at the level of gene transcription. 2. The amount of these isoenzymes can be increased by glucocorticosteroids, with the exception of PKC theta in the T cells. 3. The decreased production of arachidonic acid in the monocytes of SLE patients can be related to the inpairment of PKC delta what shows a new aspect of monocyte defects linked to this disease. 4. The absolute number of regulatory CD4+ CD25+ Fox P3+ (suppressor) T cells is decreased in the peripheral blood of SLE patients what can be increased by plasmapheresis, and what is parallel with the clinical imrpovement of the patients. 5. In HaCaT keratinocytes PKC alpha and PKC delta stimulates the differentiation and apoptosis whereas in immunodificient mice these isotypes of PKC inhibits the proliferation and tumor grouth. On the other hand, PKC beta and epsilon increases the proliferation of keratinocytes but inhibits their differentiation and apoptosis.
