Abstract

Az általunk kidolgozott MAS a BC4 x Kismis vatkana hibridpopuláció halmozott rezisztenciagéneket hordozó genotípusainak szelekciójára alkalmas. A különböző markerekkel kapott eredményeink alapján a módszer alkalmas a génhalmozott (Run1 és Ren1 gén), hordozó egyedek elkülönítésére. A multiplex PCR-el gyorsan és költségtakarékosan szelektálhatjuk ki a mindkét lisztharmat rezisztencia gént hordozó értékes egyedeket, amelyek a molekuláris vizsgálatok nélkül, hagyományos módszerekkel nem azonosíthatóak. A (Dzsandzsal kara x Laszta) x (Katta kurgán x Perlette) hibridpopulációt rezisztencia génekkel (Ren1, Run1, Rpv1) és PM QTL-ekkel kapcsolt markerekkel teszteltük. Összehasonlítottuk a Kismis vatkana és a Dzsandzsal kara rezisztencia génjeit molekuláris markerek alapján. Eredményeink azt mutatták, hogy a 13-as kapcsoltsági csoportban a Ren1 lisztharmat rezisztencia génnel kapcsolt SSR markerek rezisztenciát jelző alléljei megegyeztek, így a két rezisztencia gén azonos. Az irodalomban leírt PM QTL-lel kapcsolt markerek nem alkalmasak a populáció vizsgálatára, mert a rezisztens szülő (Dzsandzsal kara x Laszta) az SSR analízis alapján homozigóta a lisztharmat QTL-ekkel kapcsolt mikroszatellit markerekkel kapott allélokra. A PM QTL-lel kapcsolt ScORA760 SCAR markerrel jellemeztünk magyar nemesítésű rezisztens fajtákat. Eredményeik segítséget nyújthatnak a fajtaválasztáshoz egy rezisztencia nemesítési programban, ahol a különböző rezisztencia gének kombinálása, halmozása a cél. | The purpose of our study was to select the genotypes carrying pyramided resistance genes and to follow the single resistance genes in the BC4 (VRH 3082-1-42) x V. vinifera ‘Kishmish vatkana’ progenies with linked markers. Using multiplex PCR enables us to select the valuable genotypes in a single step, saving time, effort and sources. Marker-assisted selection is indispensable for selecting Run1+/Ren1+ genotypes due to the same phenotypic effect of both resistance genes. Our results demonstrate that SSR markers developed for the mapping of the disease resistance loci can be applied for MAS (marker assisted selection). We tested the (‘Dzhandzhal kara’ x ’Laszta’) x ( ’Katta kurgán’ x’Perlette’) hyibrid population with markers linked to known resistance genes (Ren1, Run1, Rpv1) and PM QTLs because of the ‘Seyve Villard’ origin of ‘Laszta’. The population enabled us to compare the resistance genes of the two Central-Asian table grape cultivars, ‘Kishmish vatkana’ and ‘Dzhandzhal kara’. The data showed that SSR profiles in Ren1 linked loci on LG 12 that ‘Kishmish vatkana’ and ‘Dzhandzhal kara’ possess the same Ren1 PM resistance gene, confirmed by the literature. PM QTLs described in ‘Regent’ cultivar were not appropriate to analyze this population, because the resistant parent (‘Dzhandzhal kara’ x ‘Laszta’) is homozygous for the alleles of the linked SSR markers. We characterized PM resistant cultivars from Hungarian bred resistant with ScORA760 SCAR marker linked to PM QTL. Our data can be useful for a resistance breeding program, where the aim is disease resistance pyramiding with MAS.
