Abstract

A női sterilitás, amely fokozatosan klónos szaporodáshoz vezet, gyakori, de kevéssé ismert tulajdonság a növénykórokozó gombák világában. Az ivarosról klónos szaporodásra történő váltás genetikai hátterének feltárására összehasonlítottuk néhány, az aszexuális szakaszban fontos fitnesz tulajdonságot kódoló gén aktivitását a Fusarium proliferatum és a F. verticillioides vad típusú törzseiben és azok ivaros szaporodásra képtelen ?MAT1-2-1 gén-diszrupciós mutánsaiban. A F. verticillioides ?MAT1-2-1 mutánsaiban mind a konídium-termelés, mind a karotinoid szintézis lényegesen csökkent a vad típusban mért értékekhez képest, és e változásokkal párhuzamosan drasztikusan gyengült a konídiálás-specifikus con10 gén, és a karotinoid bioszintézis gének(carB, carRA, carT) expressziós szintje. A fényreceptor WCC fehérjét kódoló gének aktivitása is csökkent a mutánsokban. A stressz-választ közvetítő jelátviteli útvonalak, a HOG1 MAPK, a CWIS MAPK és a cAMP-PKA útvonalak kiiktatása is teljes vagy részleges női sterilitáshoz vezetett. Ezek az eredmények azt mutatják, hogy a működőképes MAT1-2-1 gének hiányában a mutánsok nemcsak az ivaros szaporodásra való képességüket veszítik el, hanem más tulajdonságaikban is gyengébbek lesznek, ami bizonyítja, hogy a párosodási típus géneknek fontos fitnesz-funkciójuk van a gombák életciklusának ivartalan szakaszában is. | Female sterility, gradually leading to clonal reproduction is a frequently occurring, but lesser known trait in phytopathogenic fungi. To reveal the genetic background of switching from sexual to clonal reproduction, activities of selected genes with a proven role in fitness during the asexual stage of fungi were compared in the wild type strains of Fusarium proliferatum and F. verticillioides and their ?MAT1-2-1 gene disruption mutants, deprived of mating capability. Both conidiation and light-inducible carotenoid accumulation were significantly reduced in three independent ?FvMAT1-2-1 gene disruption mutants of F. verticillioides as compared to the wild type parental strain and these changes were accompanied by a drastic down-regulation of both conidiation specific (con10) and carotenoid biosynthesis genes (carB, carRA, carT) in the mutants. Expression of genes, encoding for the light receptor WCC also showed reduced activity in the mutants. Disruption of the stress response mediating signal transduction routes, including the HOG1 MAPK, the CWIS MAPK, and the cAMP-PKA pathways also led to female sterility. These findings demonstrate that mutants, lacking the functional MAT1-2-1 mating type gene have lost not only their sexual reproduction capability, but became also retarded in several fitness traits indicating that mating type genes have positive functions during the asexual phase of the fungal life cycle.
