Abstract

A rosszindulatú daganatok metasztatikus kaszkádjában elengedhetetlen a daganatok beereződése, valamint a tumorsejtek adhéziós-, mátrixbontó- és migrációs képessége. Korábbi morfológiai vizsgálatainkból kiindulva konfokális- és videomikroszkópia segítségével kimutattuk, hogy a különböző eredetű tumorsejtek motilitási morfológiája hasonló, a különbségek az adhéziók és a citoszkeleton alkotóinak dinamikájából adódhatnak. A fibroszarkóma sejtekkel végzett kétdimenziós vizsgálatok eredményei alapján leírt modell 3D rendszerben is helytálló, ráadásul másfajta tumorsejtek esetében is kimutatható. A migrációs válasz kialakulásában a citoszkeleton elemein kívül a motilitási szignál létrejöttének és lefolyásának is fontos szabályozó szerepe van. Humán melanóma esetében bebizonyítottuk, hogy az epidermális növekedési factor receptora (EGFR) és a hepatocita növekedési faktor receptora (c-Met) is érintett a tumorsejtek in vitro migrációs válaszának kialakulásában, valamint a tirozinkináz doménjeik blokkolása csökkenti a tumorsejtek in vivo metasztatizálási képességét. Angiogenezis-vizsgálatainkban leírtuk, hogy a rekombináns humán erythropoietin (rHuEPO) xenograft modellekben fokozza az endotheliális sejtek proliferációját, amely nagyobb átmérőjű erek kialakulásához vezet, és ezáltal fokozódik a daganatellenes kemo- és radioterápiák hatékonysága. Ezen felül kimutattuk, hogy nem-kissejtes tüdődaganatokban a keringő endotheliális őssejtek (EPC-k) száma korrelál a daganatos progresszióval. | Tumor-induced neoangiogenesis, adhesion, matrix degradation and migration of tumor cells are all important in the metastatic cascade of malignant tumors. Based on our previous morphologic studies about the in vitro migration of human fibrosarcoma cells we determined that the motility behavior of different types of human tumor cells is similar, the differences may be due to the dynamics of adhesions and the cytoskeletal elements. The 2D model described previously using confocal and videomicroscopy is valid in the 3D model system and applicable for all investigated tumor cells (colon carcinoma, melanoma, squamous cell carcinoma) as well. In addition to the cytoskeleton, the motility signals also have a pivotal role in the regulation of cell movement. We have demonstrated that the epidermal growth factor receptor (EGFR) and the hepatocyte growth factor receptor (c-Met) are involved in the motility signals of human melanoma cells, and blocking of their tyrosine kinase domain decreased the in vivo metastatic capacity of the cells. In our angiogenesis work we described that the administration of human recombinant erythropoietin (rHuEPO) provoked an enlarged tumoral vessel diameter caused by increased proliferation of endothelial cells, resulting in a better efficacy of chemo- and radiotherapy. Moreover, we have established a tight correlation between the bone marrow-derived circulating endothelial precursor cells and the progression of non-small cell lung cancer.
