Abstract

A mikrofabrikált keskeny fehérjecsíkok alkalmasnak bizonyultak a sejtmag mozgásának biofizikai vizsgálatára. Megfigyeltük, hogy az általánosan elfogadott nézettel szemben nem a centroszóma pozíciója határozza meg a sejt polarizációját. Publikáció előtt lévő eredményeink (Centrosome position and force map during nuclear migration in C6 glioma cells) összhangban vannak a PNAS 104(41):16182-7 2007 októberi paradigmaváltó cikkével. Az aszimmetrikus fehérjemintázatokon az előzetes várakozással szemben a sejtek nem mutattak aszimmetrikus mozgást a vizsgálati körülményeink között, ezért ezen mintázatok alkalmazását a projekt első évében befejeztük. Megfigyeltük és modelleztük szöveti sejtek sűrűségfüggő kollektív mozgását 2D-ban. Eredményeink megerősítik Prof. Vicsek Tamás és kollégái korábbi univerzális modelljeit, folytonos fázisátalakulást tapasztaltunk. Vizsgáltuk glia sejtek neuronális őssejtek fejlődésére kifejtett hatását in vitro. A retinsav sejt-sejt kommunikációban betöltött fejlődésbiológiai szerepéről nyertünk információkat. Megfigyeltük nyálmirigysejtek in vitro regenerálódását videomikroszkópia segítségével. Fluoreszcens videomikroszkópiához kapcsolódó eszközöket fejlesztettünk ki, melynek folyományaként 3 további pályázatot nyertünk el műszerfejlesztésre. Kutatási eredményeinket a Phys. Rev. E-ben és a FASEB J-ben publikáltuk, egy közleményünk bírálatra vár a Tissue Engineering c. folyóiratnál, egy további kéziratunkat 2008 elején publikáljuk. | Microfabricated narrow protein stripes were adequate for the biophysical investigation of nuclear motility. We observed that the position of the centrosome does not determine cell polarity during nuclear migration. Our results afore publication (Centrosome position and force map during nuclear migration in C6 glioma cells) are consistent with the breakthrough article published in PNAS 104(41):16182-7 in October 2007. Cells did not show asymmetric motility on the asymmetric protein patterns in our experiments denying previous expectations, so we finished these studies in the first year of the project. We observed the collective migration of cells on micropatterned surfaces as a function of cell density, and modeled the phenomenon by computer simulation. Our results confirm the previous universal model of Vicsek at al: we found a continuous phase transition. We investigated the impact of glial cells on the development of neuronal progenitor cells. We gained information of the role of retinoic acid in cell-cell communication during cell development. We observed the in vitro regeneration of human submandibular gland cells. We also developed instruments for fluorescent time-lapse microscopy, and won three additional grants for continuing such developments. We published our research results in in Phys Rev E and the FASEB J., an article is under review at Tissue Engineering, and an additional will be publihed in 2008.
