Abstract

Kimutattuk, hogy a hidrogenáz enzimreakcióban legalább egy autokatalitikus lépés található, az autokatalizátor egy enzim forma mely egy másik enzimformával hat kölcsön. Az autokatalizátor kölcsönhat az oxidált elektron akceptorral is tehát egy olyan enzimforma, melynek a disztális vas-kén kockája redukált állapotban van. Elméleti modellt fejlesztettük, mely mindezeket a kísérleti jelenségeket megmagyarázza. Kimutattuk, hogy a Hofmeister sók hatással vannak a hidrogenáz enzimreakcióra. Kozmotropikus sók a reakciót nem befolyásolják, nagy koncentrációjú kaotropikus sók igen. CD méréseket végeztünk hidrogenázokon és citokróm c4-en, hogy a hőmérséklet és sók hatására bekövetkező szerkezetváltozásokat vizsgáljuk. A citokróm c4 fehérjét sikeresen kristályosítottuk. Kimutattuk, hogy szubsztrát limitált körülmények között a hidrogenáz reakció oszcillál. Kísérleti eredményeinkből 6 publikáció született. Ezek összesített impakt faktora 21.86. 9 nemzetközi és 3 hazai konferencián tartottunk előadást. A pályázat három PhD dolgozatot eredményezett. Ősz Judit 2005-ben, Rui Miguel Mamede Branca 2008-ban, Pankotai-Bodó Gabriella pedig 2009-ben szerzett PhD fokozatot. | We have demonstrated that the hydrogenase enzyme reaction incorporates at least one autocatalytic step. The autocatalyst is an enzyme form and during the autocatalytic reaction it interacts with another enzyme form. The autocatalyst interacts with the oxidized electron acceptor as well, acordingly it is an enzyme form where the distal FeS cluster is reduced. We have developed an enzyme kinetic model to explain the observed characteristics. The salts from Hofmeister series do also affect the hydrogenase reaction. Kozmotropic salts have no effect while chaotropic salts change the kinetic characteristics. We have performed CD experiments in order to investigate how the temperature and the Hofmeister salts induce structural changes on hydrogenase and cytochrome c4. We have successfully crystallized the cytochrome c4 protein. We have discovered that under substrate limited conditions the hydrogenase displays oscillatory behaviour. We have published six papers in international journals, the sum of their impacts is 21.86. We have contributed on nine international and three national conferences. The grant resulted three PhD thesises. Judit Ősz has got the PhD in 2005, Rui Mamede Branca in 2008 and Gabriella Pankotai-Bodó in 2009.
