Abstract

Mutations of P53 tumor suppressor are associated with several human cancer. Beside of the P53 tumor supressor function, it has been recently described in yeast that P53 implicated in in transcriptional elongation. To understand the novel role of P53 in transcriptional elongation, we examined the occupancy of P53 and RNAPII at specific gene regions and we validated the interaction between P53 and RNAPII upon transcriptional elongation blockage. Interestingly, we found that P53 was associated with the examined genes, which have not been reported earlier as P53 direct targets. During the normal transcription cycle, P53 and RNAPII co-localized at the promoters and coding regions of the genes and this localization was abolished upon transcriptional elongation block. Additionally, P53 could be immunoprecipitated both with serine-5- and serine-2-phosphorylated form of RNAPII, which might implicate that P53 associates with the transcriptionally active form of RNAPII. We also showed that the transcriptional blockage-induced RNAPII degradation involved the proteasome-dependent pathway. Our results reveal a novel role of P53 during transcriptional elongation blockage, probably by the dissociation of RNAPII from DNA.
