Abstract

Eredményeink szerint élettani körülmények között a Gq/11 jelátviteli út (vélhetőleg az M3-receptorok által aktiválva) döntő szerepet játszik az acetilkolin simaizom-kontrakciót okozó hatásának közvetítésében és ezáltal a húgyhólyag működésének szabályozásában. E szignáltranszdukciós mechanizmus mellett a Rho-ROCK útvonal is hozzájárul a muszkarinerg simaizom-kontrakció kialakulásához, azonban - az eddig leírt simaizom-működésekkel ellentétben - nem a G12/13 hanem a Gi fehérjék közvetítik e hatást a receptorok felől. Ez felveti az M2-receptorok szerepét a jelátvitelben. Mivel a Rho-ROCK útvonal túlzott aktiválódását feltételezik egyes húgyhólyag-diszfunkciók (pl. overactive bladder szindróma) kialakulásában, az M2-Gi jelátviteli út új terápiás célpont lehet e betegségek kezelésében. E hipotézis tesztelésére kutatási együttműködést kezdeményeztünk Dr. Jürgen Wess munkacsoportjával (National Institute of Diabetes and Digestive and Kidney Diseases, NIH) aminek keretében többek között M2- és M3-receptor deficiens egértörzseket fogunk vizsgálni 2010 januárjában. | Our results indicate that under physiological conditions the Gq/11 signaling pathway (probably via activation of M3-receptors) plays a dominant role in the mediation of acetylcholine-induced smooth muscle contraction and control of the urinary bladder function. Furthermore, the Rho-ROCK signaling pathway is also involved in muscarinergic smooth muscle contraction, but its activation appears to be mediated by Gi (and probably M2-receptors) instead of the G12/13 proteins, which mediate Rho activation in most of the smooth muscles. Since activation of the Rho-ROCK pathway has been implicated in the pathogenesis of some urinary bladder disorders (e. g. overactive bladder syndrome) our present results identify the M2-Gi axis as a potential therapeutic target in these diseases. In order to thest this hypothesis we initiated a research collaboration with Dr. Jürgen Wess (National Institute of Diabetes and Digestive and Kidney Diseases, NIH) and in January 2010 we are going to start our experiments with mice deficient in M2- and/or M3-receptors.
