Abstract

Az RNáz L inhbitor (Rli) fehérje élesztőben esszenciális, és úgy tűnik, hogy humán sejtekben is. Szerepe élesztőben nme régóta vált ismertté. Ez a citoszolikus vas-kén fehérje specifikus szereppel bír mind a riboszomális RNS-ek, mind a riboszómák érésében. Azokban a sejtekben, amelyekben az Rli fehérje expresszióját csökkentettük, az újonnan szintetizálódó fehérjék szintje is csökkent. Ennek a jelenségnek a hátterében az állt, hogy különböző riboszomális komponensek (RNS-ek és riboszomális fehérjék) érése károsodott. Legújabb eredményeink szerint a humán RNáz L inhibitor fehérje, amelynek eddig az interferon hatásban betöltött szerepe volt ismert, szintén rendelkezik a transzlációt szabályozó nélkülözhetetlen szereppel. A humán Rli fehérje szintjének csökkentése a transzlációs szint csökkenését és a riboszomális RNS szintek változását idézte elő. | RNase L inhibitor (Rli) is an essential protein in yeast and apparently in human as well. Its function has been solved in yeast recently. This iron-sulfur protein has a specific role in the maturation of ribosomal RNAs, as well as of ribosomes. Those cells where the expression of Rli was reduced had the translation of new proteins diminished. The reason for this phenomenon was the defect in the maturation of the different ribosomal components, rRNAs and ribosomal proteins. According to our latest results the human RNase L inhibitor protein, which so far was know by its involvement in the interferon response has the same essential role in translation. Downregulation of the expression of human Rli caused reduced translation and changes in the ribosomal RNA levels.
