Abstract
This study aims to collect information about the diversity of seed storage proteins in Central European landraces and compare it to modern cultivars. The quantitative comparison of the protein composition is also monitored to demonstrate the alteration of the glutenin-to-gliadin ratio and the size distribution of the polymeric glutenins in landraces, old, and modern cultivars. In this study, 197 Central European winter wheat landraces and 67 modern winter wheat cultivars with different genetic backgrounds were examined. Employing a 20K SNP array, we explored the available genetic diversity within a collection of Central European bread wheat landraces and compared these landraces to modern wheat cultivars. For proteomic characterization of the analyzed population SE- and RP-HPLC, and MALDI-TOF analyses were carried out in order to identify new genetic sources for breeding. Comparative results were made accessible through interactive, user-friendly PCA plots, allowing efficient data exploration. Significant differences in protein content and composition were observed between landraces and modern wheat cultivars. A comprehensive analysis of the Zeleny index, grain gluten, and protein content, supported by population structure analysis using storage protein subunit markers, revealed previously untapped variation for the improvement of protein content and end-use quality in wheat breeding.
