Abstract

Molekulakönyvtárak vizsgálatára kidolgozott HPLC módszerrel összefüggéseket állapítottunk meg a szerkezet-hidrofóbitás és szerkezet-membránkölcsönhatás jellemzésére. Mért és számított lipofilitási adatok valamint biológiai aktivitás alapján egy lipofilitási tartományt határoztunk meg ahol jó antiproliferatív hatás várható. Membránokkal való kölcsönhatást immobilizált mesterséges membrán (IAM) oszlopon vizsgáltuk. Izomer vegyületeknél a tradicionális fordított fázisú oszlopnál, az IAM oszlop hatékonyabb volt a vegyületek elválasztásában, így lipofilitásuk megkülönböztetésében is. Fizikai határfelületi vizsgálati módszerekkel különböző polimerek bevonatképző tulajdonságait, a lipid réteg és hatóanyagok kölcsönhatását határoztuk meg. A falbevonat PLA/Pluronic összetételének változtatásával a nem kívánatos adszorpció csökkenthető. A micelláris elektrokinetikus kromatográfiában alkalmazott detergensekből, kevert pszeudo-állófázisok lipofilicitását és metilén-szelektivitását vizsgálva a retenciós paraméterek és a kevert pszeudo-állófázisok összetétele között lineáris összefüggést állapítottunk meg. Klinika II fázison túljutott, antitumor és neurogén gyulladásgátló hatású, TT232 heptapeptid követő molekuláinak a színtézise és karakterizálása során új, hatékony, szerkezetileg egyszerűbb, szabadalommal védett, orálisan is ható molekulákhoz jutottunk el, lehetőséget nyújtva ezzel a TT232 követő molekuláiként egy új gyógyszer kifejlesztésére. | By the aid of efficient and fast HPLC methods structure-lipophilicity and structure-membrane interactions of molecular libraries were characterized. On the bases of measured and calculated lipophilicity data and biological activity an optimal lipophilicity range was determined for good antiproliferative activity. Membrane interactions were measured on immobilized artificial membrane (IAM) column. The separation of isomeric compounds was more effective on IAM columns than on traditional RP column, thus in differentiating in their lipophilicity. Different methods were used to study the interaction of different polymers at air/solution interface and to investigate the molecular interaction between the surface layer and the compound dissolved in the subphase. The unwanted adsorption can be reduced by changing the coating PLA/Pluronic ratio. Lipophilicity and methylene selectivity of mixed pseudo-stationary phases applied in micellar electrokinetic chromatography was investigated. Good linear correlation was obtained between the measured retention parameters and the composition of the mixed phases. In the course of the synthesis and characterization of the follow-up molecules of TT232, a heptapeptide inhibiting cell proliferation and neurogenic inflammation with a completed Phase II Cinical trial, new, effective, simplified, patented and orally active molecules were identified, giving possibility for the drug development of new follow-up molecules of TT232.
